Development of Domestic CASA using the Personal Computer




Abstract
Computer-aided sperm analysis (CASA) has already developed in other country and it is currently being used widely. However, the cost of the equipment including the computer, software and microscope are too high because these equipments are sold
only as
a package. Due to this enormous financial burden, we have tried to use the currently existing PC. We are currently considering the use of software developed on our PC.
Thirty-one human semen specimens were analyzed using a standard manual method. The specimens were then videotaped and reanalyzed using two different CASA instruments (SNU system and CTS system). The videotaped specimens were analyzed by CASA for

frames for sperm concentration (CON), percent motility (MOT), and for 15 frames for kinematic variables (straight-line velocity, VSL; curvilinear velocity, VCL; linearity, LIN). The machine parameter settings for he two instruments were matched
as
closely as possible. CASA values were compared with each other for all measures and with manual results for sperm count and the percentage of motile sperm. @ES Results show: @EN 1) SNU and CTS values for CON are significantly different from
manual
measures and the linear regression reveals a low correlation. 2) SNU and CTS values for MOT are not significantly different from manual measures, but the linear regression reveals a low correlation. 3) SNU values for all parameter except LIN
(CON,
MOT,
VSL and VCL) are not significantly different from CTS values, and the linear regression reveals a high correlation. 4) SNU values for LIN are significantly different from CTS values, but the linear regression reveals a high correlation.
This study verifies the creditability of our CASA using the PC. We are currently working to upgrade the software. And we are also working on the sperm morphology analyzer using the same software.
